Maybe a stupid question, but how to exploit a resources? Do you build a mine on them and they are exploited, or what?

You exploit them through cities. You need to build a city nearby and the city will use stuff from the tile. You can also enhance SOME resources by building roads or mines. You can also destroy some (in forests and jungles) if you work on them.
